Design Notes
An important insight into the design of the construct came from the expertise of Prof. Dr. Mario Moerl, who emphasized the importance of maintaining the natural environment of the riboswitch (BBa_K4654011). This is crucial because it influences the folding and functionality of the riboswitch. We therefore made the decision to retain the spacer sequence between the promoter and the riboswitch.

The sequence we are using was created by White et. al. (2022) by creating a consenus sequence from different sequences of the riboswtich family1.
